| ProductBackground:TheAutotaxinInhibitorAssay,developedbyEchelonBiosciencesInc.,usesthefluorogenicautotaxinsubstrateFS-3.FS-3isanLPCanaloguethatisconjugatedwithbothafluorophoreandaquencher.Initsnativestatethequencherinterfereswiththefluorophore’sfluorescence.OnceautotaxincleavesFS-3,thefluorophorebecomesliberatedfromthequencher,resultinginincreasedfluorescence.Percentautotaxininhibitionisdeterminedbyreferencingnoinhibitorcontrols.Theautotaxininhibitor,BrP-LPA,issuppliedtopositivelycontrolforautotaxininhibition.TheassayisresistanttomodestconcentrationsofDMSOandisperformedinacontinuousassayformat,whichhastheadvantageofbeinglesssusceptIBLetoerrorarisingfrommis-timingtheadditionofcomponentstodifferentsamplewells.
